The F-15 aircraft were on a training flight from the US airbase at Lakenheath, Suffolk, when contact with the pilots was lost over the Cairngorm Mountains, Scottish Highlands, said spokeswoman the Grampian force.
Several police forces were notified by the American Air Force the planes were "overdue".
The spokeswoman said: "We have been notified that the planes are overdue.
"The Cairngorm area has been mentioned, but we have had no reports of any planes coming down."
A local mountain search and rescue team was also called out.
The British Ministry of Defence said the missing planes each carry a crew of two.
A spokesman confirmed: "Two aircraft are currently missing.
"They are F15s, and they are currently about one-and-a-half hours late. But we have no other details yet."
