An early Ian Keatley got Connacht on the scoreboard before Adrian Flavin and Frank Murphy crossed to help build an 18-3 half-time lead.
Murphy breached the visitor's defence early in the second-half for his second of the game - his fourth in all against Rovigo following his brace last weekend - and Andrew Farley sealed the win with 10 minutes remaining.
